As far as I know yubikey is supported by default in Gpg4win.

I always put absolute paths in the log-file parameter e.g.
c:\foo\scdaemon.log
Maybe this is why you don't see a log file?
You also need to restart scdaemon after changing the file (best to kill it with
task-manager).

Have you tried without the reader-port value? Where did you take it from?
I guess you have tried without luck without an scdaemon.conf ?

Mh, maybe some other Application blocks it. Indeed debug output should tell us
more whats going on.
When you have debug output its probably best to open an issue on
https://dev.gnupg.org
I don't think that the scdaemon maintainer reads this list and I don't know
much about the internals.
